Drug intelligence / Profile preview
**AK104 + AK117 + cisplatin + paclitaxel + 5-fluorouracil** is an investigational multi-drug combination therapy being tested for advanced and metastatic cancers, especially solid tumors such as gastric and gastroesophageal junction (GEJ) cancer[2][4]. AK104 is a bispecific monoclonal antibody targeting both PD-1 and CTLA-4, designed to enhance anti-tumor immune responses. AK117, also called ligufalimab, is a humanized IgG4 anti-CD47 monoclonal antibody that blocks CD47-SIRPα signaling, thereby promoting phagocytosis of tumor cells by macrophages and avoiding hemagglutination side effects[1]. Cisplatin, paclitaxel, and 5-fluorouracil are established cytotoxic chemotherapies used in various cancers: cisplatin is a DNA cross-linking agent, paclitaxel stabilizes microtubules to inhibit cell division, and 5-fluorouracil is an antimetabolite inhibiting thymidylate synthase for DNA synthesis. This combination leverages checkpoint blockade immunotherapy and cytotoxic chemotherapy for potential synergistic anti-tumor efficacy. Trials are ongoing to evaluate its safety and effectiveness in solid tumors[2][4].
