AK104 + bevacizumab is a combination therapy consisting of cadonilimab (AK104), a first-in-class bispecific monoclonal antibody targeting both programmed cell death protein 1 (PD-1) and cytotoxic T lymphocyte antigen 4 (CTLA-4), and bevacizumab, a recombinant humanized monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A). Cadonilimab blocks immune checkpoints PD-1 and CTLA-4 to enhance antitumor immune responses, while bevacizumab inhibits angiogenesis by preventing VEGF-A from binding its receptors, thereby reducing tumor blood supply. This combination is being investigated as first-line treatment for several cancers, including metastatic or recurrent cervical cancer and RAS-mutated or right-sided metastatic microsatellite stable colorectal cancer. The rationale for the combination is to synergistically enhance antitumor immunity while inhibiting tumor angiogenesis[1][3][5][6].
