Overview

AK112 + AK104 is a combination of two bispecific monoclonal antibodies developed for cancer immunotherapy. **AK104** (Cadonilimab/Cardonilizumab) targets both CTLA-4 and PD-1 immune checkpoints, and is approved in China for recurrent/metastatic cervical cancer; it is also in clinical trials for stomach cancer, liver cancer, lung cancer, and ovarian cancer. **AK112** (Ivosi) targets VEGF-A and PD-1 and is in late-stage trials (Phase 3) for non-small cell lung cancer and other solid tumors. The AK112 + AK104 combination is being evaluated, mostly with chemotherapy, for recurrent ovarian cancer and other malignancies in multi-center, open-label Phase 2 studies. Both antibodies re-activate anti-tumor immune responses: AK104 blocks PD-1 and CTLA-4 signaling to stimulate T-cell activity, whereas AK112 blocks PD-1 and VEGF signaling, providing immunomodulation and anti-angiogenic effects[3][4][5][7].
