Overview

This is a combination regimen consisting of **AK112 (Ivonescimab)**, a bispecific monoclonal antibody targeting PD-1 and VEGF-A; **AK117 (Ligufalimab)**, an anti-CD47 monoclonal antibody; plus the chemotherapeutic agents **carboplatin** (a platinum-based compound that causes DNA crosslinks) and **5-fluorouracil** (an antimetabolite that inhibits thymidylate synthase). AK112 is designed to simultaneously stimulate anti-tumor immunity and inhibit angiogenesis. AK117 blocks CD47 to enhance macrophage-mediated phagocytosis of tumor cells. Combining these immunotherapies with platinum-based and antimetabolite chemotherapy provides a multi-modal antitumor effect targeting the tumor microenvironment, immune evasion, and DNA replication. This regimen is in development for advanced malignant tumors, including gastrointestinal cancers, head and neck squamous cell carcinoma, and colorectal cancer[1][2][3][4][5].
