Overview

AK112 + AK127 is a combination of two monoclonal antibodies in clinical development for advanced solid tumors and hepatocellular carcinoma. AK112 (ivonescimab) is a **first-in-class bispecific monoclonal antibody** targeting both programmed death-1 (PD-1) and vascular endothelial growth factor A (VEGF-A), designed to simultaneously block PD-1/PD-L1 immune checkpoint signaling and inhibit pro-angiogenic VEGF function, thereby enhancing anti-tumor immune activity and suppressing tumor angiogenesis[2][4][5][6]. AK127 is a monoclonal antibody targeting TIGIT, an immune checkpoint receptor, aiming to further augment anti-tumor immunity by inhibiting TIGIT-mediated immune suppression[1]. Preclinical and early clinical data suggest that combining AK127 and AK112 provides synergistic anti-tumor effects by targeting complementary immune escape pathways in the tumor microenvironment[1][3]. Both antibodies are developed by Akeso Biopharma for immuno-oncology indications.
