AK112 + gemcitabine + cisplatin is a combination investigational therapy evaluated for treatment of advanced or metastatic non-small cell lung cancer (NSCLC) and other solid tumors. **AK112 (also called ivonescimab or SMT112)** is a novel first-in-class humanized IgG1 bispecific antibody targeting both **programmed death-1 (PD-1)** and **vascular endothelial growth factor (VEGF)**, designed to provide immunotherapy and anti-angiogenic effects. **Gemcitabine** is a nucleoside analog (antimetabolite) used in chemotherapy, and **cisplatin** is a platinum-based alkylating agent that damages DNA, inducing apoptosis. As a regimen, AK112 is administered intravenously with gemcitabine and cisplatin as standard cytotoxic chemotherapy agents. The combination has shown promising results in phase II and III trials, demonstrating improved objective response rates and progression-free survival in NSCLC. Ivonescimab enhances T-cell mediated immune response in the tumor microenvironment while gemcitabine and cisplatin provide cytotoxic effects against rapidly dividing tumor cells[1][5][7].
