AK112 + paclitaxel + carboplatin is an experimental three-drug combination regimen evaluated primarily for advanced non-small cell lung cancer (NSCLC). AK112 is a first-in-class, humanized IgG1 bispecific antibody targeting both programmed death-1 (PD-1) and vascular endothelial growth factor (VEGF), designed to enhance antitumor immunity and inhibit angiogenesis. Paclitaxel is a microtubule-stabilizing chemotherapeutic agent, and carboplatin is a platinum-based DNA-damaging agent. In phase II clinical trials, this combination has demonstrated promising efficacy as first-line treatment for advanced NSCLC without driver mutation, as well as for patients with EGFR mutations refractory to EGFR-TKI therapies. AK112 is developed by Akeso Biopharma, with trials funded jointly with the National Natural Science Foundation of China.
