Overview

AK117 + AK104 is a combination of two investigational monoclonal antibodies developed for cancer immunotherapy. AK117 (also known as Ligufalimab) is a novel humanized IgG4 monoclonal antibody targeting CD47, a macrophage immune checkpoint that allows tumor cells to evade immune destruction by phagocytic cells. By blocking the CD47-SIRPα pathway, AK117 promotes the phagocytosis of tumor cells and has demonstrated favorable safety features without causing significant hemagglutination or anemia[1][3]. AK104 (also known as Cadonilimab) is a bispecific antibody that targets both programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), key immune checkpoints involved in suppressing anti-tumor immunity. By simultaneously inhibiting PD-1 and CTLA-4, AK104 enhances T-cell mediated anti-tumor responses[8][1]. The combination of these agents aims to synergistically enhance anti-tumor immunity by promoting both innate and adaptive immune responses. Clinical studies have evaluated this combination in advanced solid tumors, including gastric/gastroesophageal junction adenocarcinoma, showing encouraging safety profiles and preliminary efficacy[1][5].
