This combination consists of four agents: Ligufalimab (AK117), a humanized IgG4 monoclonal antibody that blocks the CD47-SIRPα interaction to enable macrophage-mediated phagocytosis of tumor cells; Ivonescimab (AK112), a bispecific monoclonal antibody targeting PD-1 and VEGF-A to simultaneously inhibit two pathways involved in tumor immune evasion and angiogenesis; nab-paclitaxel, a nanoparticle albumin-bound formulation of the chemotherapy agent paclitaxel, and paclitaxel, a microtubule inhibitor. The combination is designed to synergistically engage the innate and adaptive immune systems alongside cytotoxic chemotherapy to treat advanced malignancies, and is being investigated in clinical trials for solid tumors, including gastrointestinal, hematological, and head and neck cancers[1][5][7].
