Overview

AK117 + chemotherapy refers to the combination therapy of **AK117 (ligufalimab)**, a humanized IgG4 monoclonal antibody targeting CD47, with one or more chemotherapy agents. AK117 binds to CD47 and blocks its interaction with SIRPα, thereby disrupting the "don't eat me" signal that cancer cells use to evade macrophage-mediated phagocytosis. This blockade enhances the immune-mediated clearance of tumor cells. Combining AK117 with chemotherapy agents, such as azacitidine or XELOX (oxaliplatin + capecitabine), has shown encouraging safety and efficacy in patients with various cancers, including advanced gastric/gastroesophageal junction (G/GEJ) adenocarcinoma and higher-risk myelodysplastic syndrome (MDS). The combination is under evaluation in several phase II clinical trials, and early results indicate enhanced anti-tumor activity and a tolerable safety profile, with common adverse events including anemia and gastrointestinal symptoms[2][3][6].
