AK129 + chemotherapy is an investigational combination therapy consisting of AK129, a humanized IgG1 bispecific antibody, and various cytotoxic chemotherapy regimens. AK129 simultaneously targets the immune checkpoint molecules PD-1 (programmed death-1) and LAG-3 (lymphocyte activation gene-3), which are often co-expressed on exhausted T cells. By co-blocking these pathways, AK129 aims to restore T-cell activation and enhance anti-tumor immune responses more effectively than single-agent checkpoint inhibitors. In clinical trials, AK129 is combined with standard-of-care chemotherapy agents—such as platinum compounds (carboplatin, cisplatin), taxanes (paclitaxel, docetaxel), and antimetabolites (pemetrexed, 5-fluorouracil)—to provide a multi-pronged attack on advanced solid tumors. This approach is being evaluated for the treatment of non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), and colorectal adenocarcinoma (CRC).
