Drug intelligence / Profile preview
Akurit-4 + Akurit-3 is a sequential combination regimen of two fixed-dose combination (FDC) products developed by Lupin Ltd. for the treatment of tuberculosis caused by *Mycobacterium tuberculosis*. Akurit-4 is used during the intensive phase of treatment and contains four first-line anti-tuberculosis drugs: rifampicin (which inhibits bacterial DNA-dependent RNA polymerase), isoniazid (which inhibits mycolic acid synthesis), pyrazinamide (which disrupts membrane potential and targets aspartate decarboxylase PanD), and ethambutol (which inhibits arabinosyltransferases). Akurit-3 is used during the continuation phase and contains rifampicin, isoniazid, and ethambutol, omitting pyrazinamide to reduce toxicity (such as hyperuricemia) while maintaining bacteriological control. This sequential regimen simplifies therapy, improves patient adherence, and helps prevent the development of drug-resistant tuberculosis strains.
