Overview

Albumin-bound paclitaxel + fluorouracil + leucovorin is a combination chemotherapy regimen investigated for the treatment of advanced gastric cancer. The regimen utilizes nab-paclitaxel, a nanoparticle formulation of paclitaxel that avoids the need for solvents like Cremophor EL, potentially reducing hypersensitivity reactions and improving delivery to tumor tissues. This is combined with 5-fluorouracil (5-FU), a pyrimidine analog that acts as an antimetabolite by inhibiting thymidylate synthase, and leucovorin (calcium folinate), which acts as a biochemical modulator to enhance the binding of 5-FU to its target enzyme. This combination has been specifically studied as a second-line treatment option for patients with advanced gastric cancer who have not previously been treated with taxanes.
