Overview

This is a multi-agent perioperative chemo-immunotherapy regimen combining albumin-bound paclitaxel, the SOX chemotherapy regimen (oxaliplatin plus S-1), and the PD-1 inhibitor sintilimab. Albumin-bound paclitaxel is a microtubule inhibitor that stabilizes tubulin to prevent cell division. The SOX regimen consists of oxaliplatin, a platinum-based DNA cross-linking agent, and S-1, an oral fluoropyrimidine combination containing tegafur (a prodrug of 5-fluorouracil), gimeracil (a dihydropyrimidine dehydrogenase inhibitor), and oteracil potassium (an orotate phosphoribosyltransferase inhibitor). Sintilimab is a recombinant humanized IgG4 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor to enhance the immune system's anti-tumor response. This specific combination is being evaluated by Peking University Cancer Hospital & Institute for the treatment of locally advanced HER2-negative hepatoid adenocarcinoma of the stomach (HAS), a rare and aggressive gastric cancer subtype.
