This is a combination therapy consisting of albumin-bound paclitaxel (nab-paclitaxel) and tigatuzumab. Albumin-bound paclitaxel is a solvent-free, nanoparticle formulation of the cytotoxic agent paclitaxel bound to human serum albumin, which enhances drug delivery to tumors via active transport mechanisms such as the gp60/caveolin-1 pathway and SPARC protein binding. Tigatuzumab is a humanized IgG1 monoclonal antibody that acts as an agonist of death receptor 5 (DR5), triggering apoptosis in DR5-positive tumor cells without requiring crosslinking. The combination has been investigated primarily in metastatic triple-negative breast cancer (TNBC), where preclinical data suggested synergistic antitumor activity due to enhanced apoptosis when both agents are used together. Clinical trials have shown that this regimen is generally well tolerated, with manageable toxicity profiles and some evidence of prolonged progression-free survival in select patients[1][2][3][4][6][8].
