Alcohol + oxycodone refers to the combined administration of ethanol and the semi-synthetic opioid oxycodone, a combination primarily studied for its pharmacodynamic interactions and abuse liability. Research led by Sharon Walsh at the University of Kentucky utilizes this combination in Phase 1 clinical trials to evaluate subjective drug effects, physiological responses, and safety profiles in individuals with a history of non-medical opioid use. Alcohol acts as a positive allosteric modulator of GABA-A receptors and an NMDA receptor antagonist, while oxycodone is a potent mu-opioid receptor agonist. The co-ingestion of these substances is of high clinical concern due to synergistic central nervous system depression, which significantly increases the risk of fatal respiratory depression and enhances the reinforcing effects that drive opioid and alcohol use disorders.
