Overview

A combination therapy of **alefacept**, **tacrolimus**, and **mycophenolate mofetil** is investigated for the prevention of allograft rejection following solid organ transplantation, particularly renal transplantation. Alefacept is a recombinant fusion protein (LFA3/IgG1) that reduces the number of memory T cells; tacrolimus is a calcineurin inhibitor that suppresses T-cell activation; mycophenolate mofetil is a prodrug of mycophenolic acid that inhibits inosine-5'-monophosphate dehydrogenase (IMPDH), critical for de novo purine synthesis in lymphocytes. Together, these agents target cellular and humoral immune responses to reduce acute rejection rates. The combination is mainly used as an immunosuppressive regimen in clinical research[1].
