Overview

This is a specific reduced-intensity conditioning (RIC) preparative regimen developed by the Masonic Cancer Center at the University of Minnesota for use in allogeneic hematopoietic stem cell transplantation (HSCT). The regimen is designed for patients with high-risk non-malignant hematologic disorders, including sickle cell disease, thalassemia, and various bone marrow failure syndromes. It consists of a combination of alemtuzumab (a monoclonal antibody targeting CD52 for lymphodepletion), cyclophosphamide (an alkylating agent), fludarabine (a purine analog that inhibits DNA synthesis), and low-dose total body irradiation (TBI). This combination aims to achieve stable donor chimerism and engraftment while reducing the treatment-related mortality and organ toxicity typically associated with full myeloablative conditioning.
