Overview

Alemtuzumab + deoxyspergualin is an experimental combination therapy consisting of two agents: - **Alemtuzumab** is a humanized monoclonal antibody targeting CD52, a glycoprotein present on the surface of mature lymphocytes. It induces profound depletion of T and B lymphocytes via antibody-dependent cellular cytolysis and complement-mediated lysis. Alemtuzumab is approved for the treatment of B-cell chronic lymphocytic leukemia and relapsing forms of multiple sclerosis. - **Deoxyspergualin (DSG)** is an immunosuppressive agent with inhibitory effects on monocytes and macrophages. This combination has been studied in renal transplantation to test whether perioperative T-cell depletion with alemtuzumab plus short-course DSG could induce immune tolerance to allografts by suppressing both lymphocyte and monocyte/macrophage activity. In clinical trials, this regimen did not achieve tolerance but resulted in reversible rejection episodes similar to those seen with alemtuzumab alone[1].
