Overview

Alemtuzumab + methylprednisolone is a combination therapy regimen evaluated by the Royal Marsden NHS Foundation Trust for the treatment of chronic lymphocytic leukemia (CLL), specifically targeting patients with p53 deletions or mutations. The regimen consists of the humanized monoclonal antibody alemtuzumab, which targets the CD52 antigen expressed on the surface of CLL cells, and high-dose methylprednisolone (HDMP), a potent corticosteroid that acts as a glucocorticoid receptor agonist. This combination is particularly significant because both agents induce apoptosis in CLL cells through mechanisms that are independent of the p53 pathway, providing a therapeutic alternative for patients who are typically resistant to conventional chemotherapy. The regimen was studied in clinical trials such as the UKCLL06 CAM-PRED trial and the National Cancer Research Institute CLL206 trial.
