Overview

Alisertib + docetaxel is a drug combination being investigated for the treatment of advanced/metastatic solid tumors. Alisertib is an oral small molecule inhibitor targeting Aurora kinase A, which is involved in the regulation of mitosis and is overexpressed in many cancers. Docetaxel is a cytotoxic chemotherapeutic drug in the taxane class that promotes microtubule stabilization and disrupts cell division. The combination aims to enhance antitumor efficacy by simultaneously disrupting mitotic spindle formation (alisertib) and stabilizing microtubules (docetaxel), leading to increased cancer cell death. This combination has shown activity in adult patients with metastatic cancers, including responses in bladder cancer, prostate cancer, and angiosarcoma. Common toxicities include neutropenia, stomatitis, and infections[1][2][4].
