Overview

Alisertib + paclitaxel is a combination therapy consisting of alisertib, an oral small molecule inhibitor of Aurora kinase A (AURKA), and paclitaxel, a microtubule-stabilizing chemotherapeutic agent. Alisertib inhibits AURKA, a serine/threonine kinase involved in mitotic progression and tumor cell proliferation. Paclitaxel disrupts mitotic spindle assembly by stabilizing microtubules. The combination is designed to synergistically impair cancer cell division and survival in rapidly proliferative tumors. This regimen has been investigated in multiple clinical trials for advanced solid tumors including high-grade neuroendocrine tumors (NETs), small cell lung cancer (SCLC), metastatic breast cancer (both hormone receptor positive/HER2-negative and triple negative subtypes), as well as ovarian cancer. Clinical studies have shown that the combination can improve progression-free survival compared to single-agent therapy with manageable toxicity profiles[1][4][5].
