Overview

**alisporivir + EDP239** is a combination of two antiviral agents under investigation for the treatment of chronic hepatitis C virus (HCV) infection. **Alisporivir** (DEB025) is a host-targeting antiviral (HTA) and cyclophilin inhibitor that blocks HCV replication by targeting host proteins necessary for viral replication and shows pangenotypic activity with a high barrier to resistance. **EDP239** is a direct-acting antiviral (DAA), specifically an NS5A inhibitor, directly inhibiting the hepatitis C virus NS5A protein, essential for viral replication. In preclinical and early clinical studies, the combination demonstrated additive to synergistic effects in suppressing HCV replication and reduced the development of resistance, supporting its development as a next-generation, interferon-free regimen for HCV[3][5][1].
