This is a combination therapy consisting of all-trans retinoic acid (ATRA; also known as tretinoin, marketed as Vesanoid) and paclitaxel (marketed as Taxol). ATRA is a derivative of vitamin A that induces cellular differentiation and inhibits proliferation by activating nuclear retinoic acid receptors. Paclitaxel is a microtubule-stabilizing agent that disrupts mitosis and induces apoptosis in cancer cells. The rationale for combining these agents stems from preclinical evidence showing that ATRA can potentiate the cytotoxic effects of taxanes like paclitaxel and reverse mechanisms of drug resistance such as epithelial-mesenchymal transition (EMT). This combination has been investigated primarily in oncology settings, including metastatic or recurrent breast cancer and other solid tumors resistant to standard therapies. Studies have shown the regimen to be relatively well tolerated with modest response rates but notable disease stabilization[1][2][4]. Nanoparticle-based co-delivery systems are also under investigation to enhance tumor targeting and reduce toxicity[3][5].
