Allisartan + benazepril is a combination therapy comprising allisartan isoproxil, a selective angiotensin II receptor blocker (ARB), and benazepril, an angiotensin-converting enzyme (ACE) inhibitor. This combination is designed to provide a comprehensive dual blockade of the renin-angiotensin-aldosterone system (RAAS). Allisartan isoproxil is a prodrug developed in China that is metabolized into the active compound EXP3174, which selectively antagonizes the angiotensin II type 1 (AT1) receptor. Benazepril inhibits the ACE enzyme, thereby reducing the conversion of angiotensin I to the potent vasoconstrictor angiotensin II. This therapeutic approach is primarily investigated for its efficacy in reducing proteinuria and slowing the progression of chronic kidney disease (CKD). Clinical research, including Phase 4 trials, has been conducted by The First Affiliated Hospital, College of Medicine, Zhejiang University to evaluate the safety and renal outcomes of this dual regimen compared to monotherapy.
