Overview

ALLO-329 + cyclophosphamide is a combination of an investigational allogeneic, CRISPR-edited dual-targeting chimeric antigen receptor T-cell (CAR-T) therapy (ALLO-329) and cyclophosphamide, a standard chemotherapy agent used for lymphodepletion. ALLO-329 is designed to target both CD19+ B cells and CD70+ activated T cells, aiming to address autoimmunity by depleting pathogenic immune cells involved in diseases such as systemic lupus erythematosus, lupus nephritis, idiopathic inflammatory myopathies, and systemic sclerosis. Cyclophosphamide is used in this combination to induce lymphodepletion, facilitating the engraftment and activity of the CAR-T cells, although ALLO-329 is engineered with Allogene’s Dagger technology to reduce or eliminate the need for lymphodepletion. The regimen is currently being studied in a Phase 1 clinical trial (RESOLUTION, NCT07085104) for adults with autoimmune diseases[1][2][3][4][5][6].
