Overview

**ALLO-605 + ALLO-647** is an investigational combination therapy being studied for relapsed or refractory multiple myeloma. **ALLO-605** is an allogeneic (donor-derived), gene-edited CAR T-cell therapy targeting **B-cell maturation antigen (BCMA)**, utilizing proprietary TurboCAR technology to increase T-cell function and resistance to exhaustion. **ALLO-647** is a humanized monoclonal antibody directed against CD52, used as part of the preconditioning (lymphodepletion) regimen to deplete host immune cells and enable persistence and effectiveness of the CAR T-cell therapy. The combination aims to provide an off-the-shelf CAR T-cell option with improved safety and potency, particularly in patients who have failed previous multiple myeloma therapies. Both are being developed by Allogene Therapeutics and are in phase 1 clinical evaluation[1][2][3][4][5].
