Allo-RevCAR01-T + R-TM123 is a combination immunotherapy drug under investigation for the treatment of CD123-positive hematologic malignancies, notably acute myeloid leukemia. It consists of two components: "Allo-RevCAR01-T," which is an allogeneic human T-cell therapy genetically modified to express a reversed, universal chimeric antigen receptor (RevCAR) presenting an extracellular peptide epitope, and "R-TM123," a recombinant antibody derivative acting as a bridging molecule. R-TM123 specifically binds both the RevCAR epitope on the T cell and the CD123 antigen expressed on the target cancer cell, allowing targeted cytotoxicity. The activity and cytolytic response of the CAR-T cells is strictly dependent on the presence of R-TM123, thus enabling rapid switchability in case of adverse events. This system is designed to improve safety and provide controllable, potent anti-leukemic efficacy[1][3][5][6][7].
