Overview

Allocetra-OTS + tislelizumab is a combination of two investigational immunotherapeutic agents being studied for the treatment of advanced-stage solid tumors[1][2][3][4][6]. **Allocetra-OTS** is a universal, off-the-shelf cell therapy composed of allogeneic peripheral blood mononuclear cells, engineered to induce *reprogramming of macrophages into their homeostatic state*, thereby restoring immune homeostasis and potentially rebalancing the immune system in life-threatening diseases such as cancer and sepsis[2][3][6]. **Tislelizumab** is a humanized IgG4 anti–PD-1 monoclonal antibody immune checkpoint inhibitor designed to minimize binding to the Fcγ receptor on macrophages, thereby avoiding antibody-dependent T-cell inhibition and improving anti-tumor immune responses[2][3][4]. The combination aims to synergistically enhance anti-tumor immune responses by reprogramming macrophages (via Allocetra-OTS) while simultaneously blocking the PD-1 immune checkpoint pathway (via tislelizumab)[2][4]. Allocetra-OTS is being developed by Enlivex Therapeutics, and tislelizumab is developed and supplied by BeiGene[1][2][3][4][5]. This combination is currently under investigation in a Phase I/II study in patients with advanced solid tumors who have failed previous anti-PD-1 therapies[1][2][3][4][6].
