Overview

This is an investigational allogeneic, third-party CD19-specific CAR T-cell therapy developed by M.D. Anderson Cancer Center for the treatment of refractory CD19-positive B-lineage lymphoid malignancies. The therapy consists of donor-derived T cells that are genetically modified using the non-viral Sleeping Beauty transposon system to express a chimeric antigen receptor (CAR) targeting the CD19 antigen. The CAR construct typically incorporates CD28 and CD3-ζ signaling domains to facilitate T-cell activation and proliferation upon binding to malignant B cells. Unlike autologous CAR T-cell therapies, these cells are sourced from unrelated third-party donors and are selectively expanded using artificial antigen-presenting cells (aAPCs). The therapy is primarily evaluated in Phase 1 clinical trials for patients with relapsed or refractory B-cell leukemia or lymphoma, particularly those who have failed prior stem cell transplantation.
