Allogeneic CD8+ memory T-cells are a specialized population of donor-derived cytotoxic T-lymphocytes used in adoptive immunotherapy, primarily following allogeneic hematopoietic cell transplantation (HCT). This cell therapy involves the purification and infusion of donor-derived CD8+ T-cells that exhibit a memory phenotype (e.g., central memory or effector memory cells), typically characterized by the expression of markers such as CD45RO and the absence of CD45RA. The therapeutic objective is to induce a robust graft-versus-leukemia (GVL) or graft-versus-tumor effect by targeting minor histocompatibility antigens (mHAs) or tumor-associated antigens (TAAs) on residual malignant cells. By specifically selecting memory T-cells, this approach aims to maintain potent anti-tumor activity while potentially reducing the risk or severity of graft-versus-host disease (GVHD) compared to traditional unselected donor lymphocyte infusions (DLI). This strategy is often employed as a consolidative therapy after non-myeloablative conditioning to eradicate minimal residual disease in various hematologic malignancies.
