Overview

Allogeneic HPV-specific CD4+ T cells are an experimental adoptive cell therapy developed by the Sidney Kimmel Comprehensive Cancer Center at Johns Hopkins. The therapy is designed for patients with advanced HPV16-associated malignancies, such as head and neck squamous cell carcinoma. The production process involves vaccinating a haploidentical relative with a therapeutic HPV16 vaccine to induce a specific immune response. Peripheral blood cells are then harvested from the donor and depleted of CD8+ T cells to minimize the risk of graft-versus-host disease (GvHD) while retaining HPV-specific CD4+ helper T cells. These cells are subsequently infused into the patient to coordinate an anti-tumor immune response against HPV16-positive cells.
