This investigational adoptive cellular therapy, developed by Tufts Medical Center, consists of haploidentical allogeneic natural killer (NK) cell-enriched mononuclear cells. The therapy utilizes CD3-depleted, MHC-mismatched NK cells obtained from related donors via apheresis. These cells undergo immunomagnetic CD3 depletion to minimize the risk of graft-versus-host disease (GvHD) and are expanded ex vivo with interleukin-2 (IL-2) and human AB serum. The product is administered intravenously to patients with hematological malignancies, including acute myeloid leukemia, non-Hodgkin lymphoma, Hodgkin lymphoma, and multiple myeloma, specifically following an autologous stem cell transplant (ASCT). The mechanism of action relies on the MHC-independent cytotoxicity of NK cells to identify and eradicate residual malignant cells that may have survived prior high-dose chemotherapy or radiation.
