This experimental combination immunotherapy involves the sequential administration of allogeneic natural killer (NK) cells and allogeneic dendritic cells (DCs) to treat advanced malignancies, specifically recurrent glioblastoma. The protocol utilizes NK cells derived from a haploidentical donor, which are activated ex vivo with Interleukin-2 (IL-2) to enhance their innate cytotoxic potential against tumor cells. This is followed by the administration of monocyte-derived dendritic cells pulsed with the CMV-pp65 antigen, a protein frequently expressed in glioblastoma tissues. The dual mechanism aims to trigger immediate innate immune destruction of cancer cells via NK cells while simultaneously priming the adaptive immune system through DC-mediated T-cell activation against specific tumor-associated antigens. This approach is designed to overcome the highly immunosuppressive environment of the central nervous system and has demonstrated the ability to induce complete clinical responses in individual case studies.
