Overview

Allogeneic NK cell therapy (The Second Hospital of Shandong University) is an adoptive immunotherapy being investigated for the treatment of elderly patients with recurrent or refractory acute myeloid leukemia (AML). The therapy involves the infusion of natural killer (NK) cells derived from a healthy allogeneic donor. In clinical study NCT04324190, these cells are administered as part of a multi-agent regimen that includes lymphodepleting chemotherapy with cyclophosphamide, priming with G-CSF, and the hypomethylating agent azacitidine to enhance anti-leukemic activity. NK cells exert their therapeutic effect by recognizing and eliminating malignant cells through the release of cytolytic granules containing perforin and granzymes, as well as through the activation of death receptor-mediated apoptosis, providing a graft-versus-leukemia effect without the high risk of graft-versus-host disease associated with T-cell therapies.
