Overview

An allogeneic Vγ9Vδ2 T cell immunotherapy being developed by Beijing Tiantan Hospital for the treatment of malignant gliomas, including glioblastoma multiforme (GBM) and diffuse intrinsic pontine glioma (DIPG). These cells are a subset of gamma-delta T cells that recognize phosphoantigens, such as isopentenyl pyrophosphate (IPP), which accumulate in glioma cells due to dysregulated cholesterol metabolism. This recognition is mediated by the butyrophilin molecules BTN3A1 and BTN2A1 and occurs independently of the major histocompatibility complex (MHC), facilitating allogeneic transplantation without the need for HLA matching. The therapy is administered locally into the cerebral ventricles via an Ommaya capsule to bypass the blood-brain barrier and directly target the tumor site.
