This experimental immunotherapy protocol, developed by the Centre Hospitalier Universitaire de Besançon, involves the adoptive transfer of allogeneic, haploidentical natural killer (NK) cells in combination with the monoclonal antibody cetuximab. Specifically designed for patients with EGFR-positive metastatic gastrointestinal cancers and liver metastases, the NK cells are administered via hepatic intraarterial infusion to achieve high local concentrations in the liver. The treatment regimen includes lymphodepleting conditioning with cyclophosphamide and fludarabine, followed by the NK cell infusion, weekly cetuximab to facilitate antibody-dependent cellular cytotoxicity (ADCC) against tumor cells, and interleukin-2 (IL-2) to support the survival, expansion, and activation of the transferred NK cells.
