ALMB-0168 + PD-1 inhibitor is a preclinical combination therapy being developed by AlaMab Therapeutics for the treatment of solid tumors and bone-involved cancers. The combination consists of ALMB-0168, a first-in-class humanized monoclonal antibody that targets Connexin 43 (Cx43) hemichannels, and an unspecified PD-1 (programmed death-1) checkpoint inhibitor. ALMB-0168 acts as an agonist to activate Cx43 hemichannels, which triggers the release of ATP and other tumor-inhibiting factors into the tumor microenvironment. This mechanism is intended to work synergistically with the PD-1 inhibitor, which enhances T-cell mediated anti-tumor immunity. The program is currently in the IND-enabling or early clinical development stage for bone-involved cancers.
