Almonertinib + carboplatin + pemetrexed is a combination regimen used in the treatment of non-small cell lung cancer (NSCLC), particularly in patients with EGFR mutations or after failure of prior EGFR tyrosine kinase inhibitor therapy. Almonertinib is a third-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor that selectively targets both EGFR-sensitizing and T790M resistance mutations, inhibiting tumor cell proliferation. Carboplatin is a platinum-based chemotherapeutic agent that causes DNA crosslinking and apoptosis in rapidly dividing cells. Pemetrexed is an antifolate chemotherapy drug that inhibits multiple folate-dependent enzymes involved in nucleotide synthesis, thereby preventing DNA replication and cell division. The combination aims to enhance antitumor efficacy by targeting cancer cells through different mechanisms: targeted inhibition of mutant EGFR signaling, DNA damage via platinum compound, and disruption of nucleotide synthesis[3][5][7].
