Almonertinib + pyrotinib is an investigational combination therapy consisting of a third-generation EGFR tyrosine kinase inhibitor (almonertinib) and an irreversible pan-ErbB receptor tyrosine kinase inhibitor (pyrotinib). This regimen is primarily being evaluated for the treatment of patients with locally advanced or metastatic non-small cell lung cancer (NSCLC) who have developed resistance to prior EGFR-TKI therapy and harbor HER2 amplification or mutations. Almonertinib selectively targets EGFR mutations, including T790M, while pyrotinib inhibits HER1, HER2, and HER4, effectively blocking downstream signaling pathways that drive tumor growth. The combination aims to overcome resistance mechanisms mediated by HER2 alterations and is currently being studied in Phase II clinical trials.
