Drug intelligence / Profile preview
Alnodesertib plus gemcitabine is an investigational combination regimen consisting of alnodesertib (formerly ART0380), an oral, highly selective small-molecule inhibitor of ataxia telangiectasia and Rad3-related (ATR) kinase, and gemcitabine, a nucleoside analog chemotherapy. Developed by Artios Pharma, this combination is designed to exploit DNA damage response (DDR) vulnerabilities in cancer cells. Gemcitabine induces DNA damage by inhibiting DNA synthesis, while alnodesertib blocks ATR-mediated DNA repair and replication stress signaling, leading to synergistic tumor cell death. The combination has shown clinical efficacy in a randomized Phase 2a study for platinum-resistant high-grade serous ovarian cancer (HGSOC), meeting its primary endpoint of improved progression-free survival compared to gemcitabine alone.
