Alpelisib + everolimus is a combination therapy targeting the PI3K/AKT/mTORC1 signaling pathway. Alpelisib is a selective inhibitor of the phosphatidylinositol 3-kinase (PI3K) alpha subunit (p110α), while everolimus is an mTORC1 (mechanistic target of rapamycin complex 1) inhibitor. This combination provides synergistic efficacy by simultaneously blocking two critical points in the PI3K/AKT/mTORC1 pathway, which is implicated in cancer progression and resistance to anti-estrogen treatments.
