Alpelisib + ganitumab is an investigational **combination therapy** composed of **alpelisib**, a selective inhibitor of the phosphatidylinositol 3-kinase catalytic subunit alpha (PI3Kα), and **ganitumab**, a fully human monoclonal antibody targeting the insulin-like growth factor-1 receptor (IGF-1R). - **Alpelisib** inhibits the PI3K/AKT/mTOR pathway, commonly activated in cancer through **PIK3CA mutations**, to decrease tumor growth and proliferation. - **Ganitumab** binds and blocks IGF-1R, inhibiting receptor-mediated signaling involved in cell survival and proliferation. This combination is under investigation for cancer indications where these pathways are implicated in disease progression or drug resistance.
