Overview

Alpelisib (BYL719) and olaparib are two small molecule drugs being studied in combination for the treatment of various cancers, particularly platinum-resistant or refractory high-grade serous ovarian cancer without germline BRCA mutations. Alpelisib is a selective inhibitor of phosphatidylinositol 3-kinase alpha (PI3Kα), which plays a key role in the PI3K/AKT signaling pathway involved in cell growth and survival. Olaparib is a poly(ADP-ribose) polymerase (PARP) inhibitor that disrupts DNA repair processes, leading to tumor cell death, especially in cells with defective homologous recombination repair mechanisms. The combination aims to exploit potential synergistic effects by targeting both DNA repair and cell survival pathways[1][2][4][8].
