Alpelisib (BYL719) and olaparib are two small molecule drugs being studied in combination for the treatment of various cancers, particularly platinum-resistant or refractory high-grade serous ovarian cancer without germline BRCA mutations. Alpelisib is a selective inhibitor of phosphatidylinositol 3-kinase alpha (PI3Kα), which plays a key role in the PI3K/AKT signaling pathway involved in cell growth and survival. Olaparib is a poly(ADP-ribose) polymerase (PARP) inhibitor that disrupts DNA repair processes, leading to tumor cell death, especially in cells with defective homologous recombination repair mechanisms.
