alpha 2,3 sialyltransferase-IN-1 is a small molecule inhibitor of alpha 2,3-sialyltransferase (ST3Gal), an enzyme that catalyzes the transfer of sialic acid to the terminal positions of glycan chains on proteins and lipids. In the context of Alzheimer's disease (AD) research, this compound is utilized to modulate the glycosylation patterns of neutrophils. Elevated sialic acid residues on neutrophil membranes have been linked to increased vascular adhesion and the formation of capillary stalls, which contribute to cerebral hypoperfusion and cognitive decline. By reducing these sialic acid caps, alpha 2,3 sialyltransferase-IN-1 has demonstrated the ability to improve cerebral blood flow and reduce capillary obstruction in amyloidosis mouse models. This research, primarily conducted at the University of Miami, suggests that targeting aberrant sialylation may offer a novel therapeutic strategy for mitigating the vascular components of AD pathogenesis.
