Bionomics is developing small molecule negative allosteric modulators (NAMs) of the alpha7 nicotinic acetylcholine receptor (α7 nAChR) for the treatment of various central nervous system (CNS) disorders. Unlike positive allosteric modulators (PAMs) which enhance receptor activity, NAMs reduce the receptor's response to its endogenous ligand, acetylcholine. This mechanism is being explored for conditions where overactivity of the α7 nAChR is implicated. The program is currently in the preclinical lead candidate stage and is part of Bionomics' broader cholinergic platform.
