Drug intelligence / Profile preview
Alpitatug satetraxetan + pasritamig is an investigational combination of two human kallikrein-2 (KLK2)-targeted agents developed by Johnson & Johnson (Janssen) for the treatment of advanced prostate cancer, specifically metastatic castration-resistant prostate cancer (mCRPC). Alpitatug satetraxetan (JNJ-69086420) is a first-in-class radioimmunoconjugate consisting of a humanized IgG1-kappa monoclonal antibody targeting KLK2, conjugated to the DOTA-derivative chelator satetraxetan and radiolabeled with the alpha-emitting radionuclide actinium-225. Pasritamig (JNJ-78278343) is a first-in-class bispecific IgG1 antibody and T-cell engager that simultaneously binds KLK2 on prostate cancer cells and CD3 on T cells, redirecting T-cell-mediated cytotoxicity to the tumor. This combination is being evaluated in Phase 1 clinical trials to assess safety, tolerability, and preliminary efficacy in patients with mCRPC.
