Overview

Alrizomadlin (APG-115) is a novel, orally active small-molecule inhibitor of MDM2 that activates p53-mediated apoptosis in tumor cells and functions as a host immunomodulator. Pembrolizumab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), used as an immune checkpoint inhibitor. The combination of alrizomadlin and pembrolizumab is being investigated for the treatment of advanced solid tumors, including unresectable or metastatic melanoma, non-small cell lung cancer (NSCLC), liposarcoma, urothelial carcinoma, malignant peripheral nerve sheath tumor (MPNST), and ATM-mutant solid tumors that have progressed on prior immunotherapy. Alrizomadlin restores TP53 function in wild-type TP53 or MDM2-amplified tumors and may enhance antitumor immunity when combined with PD-1 blockade[1][2][3][4][5].
