Overview

ALT-836 + gemcitabine is a combination therapy under investigation for the treatment of locally advanced or metastatic solid tumors. ALT-836 is a recombinant human-chimeric monoclonal antibody that acts as a direct antagonist of tissue factor (TF), which is overexpressed in many cancer types and plays a key role in cancer-associated thrombosis, tumor growth, angiogenesis, and metastasis. By blocking TF activity on cancer cells, ALT-836 aims to inhibit these processes. Gemcitabine is a nucleoside analog used as standard chemotherapy for various cancers; it inhibits DNA synthesis by incorporating into DNA strands during replication. The combination seeks to leverage the anti-tumor effects of both agents—targeted inhibition of TF-driven pathways by ALT-836 and cytotoxicity from gemcitabine—to improve clinical outcomes in patients with advanced malignancies[1][2].
