Overview

The combination of ALVAC-HIV vCP1521 and AIDSVAX is a "prime-boost" HIV vaccine regimen designed to prevent human immunodeficiency virus (HIV) infection. The regimen consists of two components: ALVAC-HIV vCP1521, a recombinant canarypox viral vector vaccine developed by Sanofi Pasteur, which carries HIV genes (env, gag, and part of pol from the CRF01_AE subtype), and is non-replicative in human cells. It acts as an immunostimulant by expressing these HIV proteins to elicit an immune response[2][5]. AIDSVAX B/E is a bivalent subunit vaccine containing recombinantly expressed envelope proteins (gp120) from two strains of HIV, originally developed at Genentech and later managed by VaxGen and Global Solutions for Infectious Diseases (GSID)[5][8]. This combination was evaluated in the RV144 phase III trial in Thailand, where it demonstrated modest efficacy (~31%) in preventing HIV acquisition over six months[3][4][7]. The prime/boost approach with these vaccines induces both cellular and humoral immune responses superior to protein-only regimens[3][4].
